【摘要】 目的 研究中药姜香贴剂对离体鼠皮的透过性。方法 用ficks扩散装置,测定姜香贴剂中6-姜酚在大鼠皮肤的透皮速率,研究比较甘油、乙醇、氮酮和聚丙烯酸酯压敏胶对6-姜酚透皮速率的影响。结果 处方③20 h的累积渗透量(q)为585.898 μg/cm2,渗透系数(kp)为42.25 cm-2·h-1/2,透皮速率(js)为16.711 μg·cm-2·h-1/2,明显好于其他5个处方。结论 姜香贴剂可透过离体鼠皮,氮酮是影响6-姜酚透皮速率的重要因素。

[abstract] objective to study the permeation of the patches jiangxiang and its permeation mechanism.methods the in vitro penetration model was designed to investigate 6gingerols permeability in rat,with improved fick’s diffusing and to study the influence factors(alcohol,glycerol and azone).results the percutaneous adsorption speed of the patches jiangxiang were measured.the accumulative permeation quantity(q)was 585.898 μg/cm2,steady percutaneous speed(js) was 16.711 μg·cm-2·h-1/2and permeation coefficient(kp)was 42.25 cm-2·h-1/2.conclusion the percutaneous adsorption speed of the patches jiangxiang is passive diffusion and azone is important influence factors of the 6gingerols permeability.

姜香透皮贴剂是由生姜、广藿香和木香的超临界co2提取物制成,对旋转刺激诱发大鼠的运动病模型有良好的抑制和缓解作用。本文用ficks扩散装置[1],以6-姜酚为检测指标,通过计算6-姜酚对大鼠皮肤的累积渗透量(q)、透皮速率(js)和渗透系数(kp),研究姜香贴剂的透皮吸收行为。
1 仪器与试药
高效液相色谱仪(agilent 1050 series,美国惠普公司);紫外检测器(uv2100型,日本岛津公司);全自动体外透皮扩散实验仪(美国汉森科技公司);电子天平(bs-110s型,北京赛多利斯天平有限公司);涂布机(北京斯达特机电公司);生姜(山东莱芜);6-姜酚(中新药业,经1hnmr、13cnmr、ir、uv、ms鉴定为6-姜酚,经hplc法检查纯度>98%);氮酮(azone)(石家庄虎头山试剂厂);水为重蒸水;甲醇为色谱纯;其他试剂均为分析纯;雄性大鼠(中国科学院实验动物繁殖厂)。
